Samsung LED LCD Calibration

Hi All, I have a Samsung UN46B6000 LED LCD display and I'm getting ready to calibrate it using the Spears and Munsil disk. Where should the backlight be set before I begin? This setting is independent of the control used for setting white. I can't find any info on this. Does it matter? In other words will it calibrate to the proper picture regardles of where this setting starts? Thanks, Paul
